IN VITRO and IN VIVO STUDIES
Joint Venture of Toxindia
Mahesh Sco., Bibwewadi, Plot no.52, Nilesh Park, Pune – 411037, & Govt, College of Pharmacy, Karad & TOXINDIA, Maharashtra, India.
The study of Pinak as anti snake venom is divided into five sub studies as follows;
  • In vitro efficacy study of Pinak against Cobra venom using human blood cells.
  • In vivo efficacy study of Pinak in mouse model against Cobra venom.
  • In vivo efficacy study of Pinak in mouse model against Viper venom.
  • In vitro efficacy of Pinak against viper venom haemorrhage-line activity in mice liver.
  • Ex vivo efficacy study of Pinak against Cobra venom on isolated rat phrenic nerve diaphragm
    1. COMPARISON BETWEEN % HAEMOLYSIS OF PINAK AND SNAKE VENOM ANTISERUM
    2. In vivo efficacy, study of Pinak in mouse model against Cobra venom Pinak (Oral tablet) was foud to be effective against cobra venom when given 1 hour before and after envenomation; as a potent antivenin at the dose 300mg/kg (20mg/kg APL) (Human therapeutic dose 1.4 mg/kg).
    3. In vivo efficacy study of Pinak in mouse model against Viper venom Pinak (Oral tablet) was found to be effective against viper venom when given 1 hour before and after envenomation; as apotent antivenin at the dose 300mg/kg.
    4. In vitro efficacy of Pinak against Viper venom haemorrhage-like, activtity in mice liver in this assay the observed haemorrhage may be combination of three major factors like rupture of capillary membranes carrying blood, fibrinolytic activity to melt clots and inhibition of one or more blood clotting factors. Haemoglobin released from liver was a time dependent phenomenon followed the Second-order Kinetics as evidenced by linear dependency of varying venom concentration.
Inhibition of haemorrhagic potential of venom toxins by pinak herbal complex preparation may be considered as a preliminary drug of its anti-venom character.
Haemoglobin released (Second-order Kinetics) from the liver of BALB/c mouse was found to be arrested by Pinak herbal preparation against viper venom, indicated that Pinak herbal preparation in potent preparation against haemorrhagic action of viper venom.
      • Ex vivo efficacy study of Pinak against Cobra venom on isolated rat phrenic nerve diaphragm
Pinak at the concentration of 7 mg/ml in isolated phrenic nerve diaphragm preparation showed effective in elevating neuromuscular transmission activity when blocked by the cobra venom.
    CLINICAL STUDIES
    PINAK A STUDY AT KARAD
    • Continuous close & frequent observation is needed.
    • Keep the stomach empty till patient is out of danger.
    • We administered medicine after toxicity is well developed. It saves much antivenin. Non-toxic cases do not need any antivenin. *Swelling gets relieved much faster than injection ASV
    • First aid is very important is snakebite cases.
    • Observation of the patient for 1 hour after tourniquet is released is very important
    • A physician should treat the patient with confidence. If he is afraid, patient well definitely suffer.
    • One PINAK tablet is equivalent to about 5 ASVs.
COMPARATIVE STUDY OF PINAK WITH ASV IN SNAKEBITE IN SASOON HOSPITAL, PUNE
    • No. of ASV vials requirement was less in Pinak group than control group. P<0.05.
    • In neuroparalytic snakebite no significant different was foud but in ASV requirement, but in vasculotoxic snakebite statistical significant difference was found in ASV requirement.
    • Complications are minimal, of less severity, in Pinak group as compared to control group.
    • Neuroparalytic patients recover earlier with Pinak than ASV
A COMMUNITY TRIAL OF PINAK AT NARODI.
Pinak was kept at 11 centres in village, each centre was functional for 24 hours and Manned by somebody, pinak was given free to all, as soon as patient reaches Centre one pinak is given is given sub lingually to patient, one more pinak tablet is given along with the relative to administer if required.’
    OBSERVATION:
    • Among the admitted patients, it was observed that, administration of PINAK tablets not only prevents death but also reduces swelling, pain, bleeding
    • Recovery time also reduced significantly.
NOTE ON ROAD DEATHS WERE PREVENTED COMPLETELY WITH A SINGLE PINAK TABLET.
USE OF “PINAK” IN THE TREATMENT OF SCORPION STINGS
Pinak is a an ayurvedic antivenin used for snake bite cases. Feedback from patients of scorpion stings stimulated us for this study. Now we have concluded that a single tablet of pinak is able to cure scorpion stings cases completely. More tablets may be required in few selected cases of scorpion stings.
